An electrochemical genotyping by using two ferrocene/isoquinoline-connected DNA probes with different redox potentials on a single electrode was investigated. A solution of 100μm Iq*-ADH7 phosphate buffer was placed on a commercially available gold electrode, which was cleaned. The electrode was kept in a closed container under high humidity for 90 mm at room temperature. After rinsed with a buffer solution, the gold electrode modified with DNA probes was soaked in a 1 mM 6-mercaptohexan 1-0l solution of a buffer solution for 90 mm at room temperature, and then thoroughly washed with Milli-Q water and the buffer solution successively. The mismatched duplex Iq*1-2 was found to reveal an enhanced bending elastically compared with the matched Iq*1-1 as the duplexes from Iq1 did. This electrochemical genotyping could be conducted accurately and rapidly by using the two DNA probes, which possess different redox potentials.
